The Transponder is a Resistance device attached to a supply crate. It allows the crate to be easily found with a radar. At the start of the chapter Under The Radar, the Muscle Car is equipped with a radar in a Resistance outpost located near the Combine Autogun in the Outlands to help Gordon and Alyx find Resistance supplies equipped with transponders along the road.
The Transponder is also involved in the "Cache Checker" Orange Box achievement, involving the finding of every cache.
